Is it possible to build it like this:
I put 50 emerald blocks in a funnel, the blocks are removed and after that I have constant speed 1.
Is this possible and if so how can I build it?

You could try to build a binary counter, or if you are lucky you can solve it with an observer and a container filled with the blocks. For permanent speed you would have to give yourself per comandblock speed for 99999 seconds:-)
Simply an item sorter with counter (see Youtube), and after 50 items it triggers a signal, which triggers a command block with / effect @p minecraft: speed 99999 1.
